​Microscopy & Imaging Facility

The Microscopy and Imaging Facility is a shared resource with full access to the Lurie Children’s research community. Our researchers have unlimited access to the latest and best technological tools. The facility also provides technical support and training on our instruments, including light and fluorescent microscopes, confocal laser scanning microscopes, digital image processing and analysis and darkroom facilities.

Instrumentation

Light and Fluorescent Microscopes

Leica DMR-HC Upright Research Microscope
  • A research quality upright microscope with brightfield, differential interference contrast (DIC) and epi-fluorescent capability for blue, green, red and near ultraviolet excitation.
  • Objectives from 1X through 100X are available.
  • Uses a QImaging Retiga 4000R camera with 2048x2048 pixel resolution for grayscale and real color image capture.
Leica DM-IRB Inverted Research Microscope
  • A research quality inverted microscope with brightfield, differential interference contrast (DIC), phase contrast and epi-fluorescent capability for blue, green and near ultraviolet excitation.
  • Objectives from 5X through 100X are available.
  • Uses a QImaging Retiga 4000R camera with 2048x2048 pixel resolution for image capture.

Confocal Laser Scanning Microscopes

Zeiss LSM 880 Confocal Laser Scanning Microscope with Airyscan
  • The instrument is based on an Axio Observer inverted microscope. 
  • Seven laser excitation lines available from 4 lasers: 405, 458, 488, 514, 561, 594 and 633nm. 
  • Five detectors, three for normal confocal imaging, one for transmitted light imaging, and the Airyscan detector. 
  • Airyscan is a new confocal detector scheme that effectively captures light normally rejected by the pinhole in confocal imaging.  This extra signal is then used to improve speed, sensitivity and/or resolution. The Airyscan GaAsP detector consists of 32 detector elements that are arranged to capture the entire point spread function, or “Airy Disk” in 32 separate images.  Processing the data in these 32 images can result in an increased optical resolution of up to 1.7X in lateral (x-y) and axial (z) axes. 
  • Objectives: Plan-Neofluar 10x/0.30, LD Lci Planapo 25x/0.8 (long working-distance, multiple immersion), F-Fluar 40x/1.30 Oil, C-Apochromat 40/1.2 Water, Plan-Apochromat 63x/1.40 Oil, C-Apochromat 63x/1.20 Water and Alpha-Plan-Apochromat 100x/1.46 Oil. 
  • All objectives have differential interference contrast imaging capability. 
  • The instrument is equipped with a motorized scanning stage with controller, heated stage inserts and an objective heater for short-term live-cell imaging. 
  • The software is Zeiss ZEN 2.1 (Black edition) on a Windows-based computer system.
Zeiss LSM 800 Confocal Laser Scanning Microscope
  • A three channel confocal microscope specifically designed for long-term, live-cell imaging. Two GaAsP-PMT detectors have twice the quantum efficiency of previous PMTs which makes them much more light sensitive. This means using less laser intensity and less detector power to achieve brighter images with less fading and/or specimen damage than before. The third detector is a transmitted light detector.
  • Based on an Axio Observer Z1 inverted microscope. 
  • Four laser excitation lines from four solid-state lasers: 405, 488, 561 and 640nm. 
  • Objectives: Plan-Neofluar 2.5x/0.075, Plan-Neofluar 10x/0.30, LD Lci Planapo 25x/0.8 (long working-distance, multi immersion), LD C-Apochromat 40x/1.10 W (long working-distance, water immersion), LD C-Apochromat 63x/1.15 Water and Alpha-Plan-Apochromat 100x/1.46 Oil. All objectives have differential interference contrast imaging capability.  
  • The instrument is equipped with a motorized scanning stage. 
  • Has the Zeiss Definite Focus controller for ensuring precise focus during long-term time-lapse conditions when focus drift is often a problem. 
  • Enclosed in a full incubation system which enables precise temperature for the entire instrument and a heated stage insert for temperature control and CO2 levels at the specimen itself. 
  • Uses the Zeiss ZEN 2.1 (Blue version) Windows-based computer system. There is a software module for advanced positions and tiling and a complete analysis module for quantitative analysis. 

Image Analysis

Digital Darkroom
  • A Windows-based computer system with the Adobe Creative Suite, Adobe Acrobat and Microsoft Office Professional as the main software components.
  • The computer system is also equipped with an Epson Perfection V800 Photo scanner with Silverfast Ai scanner software.
  • Archive-quality photographs up to 17x22 inches from digital files are produced on an Epson Stylus Pro 3880 printer.
Darkroom
The facility also supports a complete "wet" darkroom containing a QPS x-ray developer for processing of films for biochemical and molecular analysis.
